Rapid Multiplication of Potato (Solanum taberosum L.) Stem Cuttings Based on Nutrient Culture System



Abstract
An efficient system for rapid multiplication of seed potatoes using a stem cutting technique has been developed by nutriculture method. The system consists of two stages: 1st, in vitro culture of plantlets for mother plant production and 2nd, nutriculture of the mother plants for rapid multiplication of stem cuttings. The growth, including rooting percentage, stem length, shoot weight, root weight and dry matter content, of mother plants for stem cuttings was good when the mother plants were produced from plantlets cultured in vitro on MS basal medium supplemented with 2¡­3% of sucrose. In vitro plantlets grown on this medium showed 100% of survival rate at 2 weeks after transplanting in the nutriculture bed. The increased concentration of nitrate nitrogen in the nutriculture solution stimulated the growth of main stems of mother plants. More number of cuttings were produced with the cutting at the apical portion of mother plants as compared with the cutting at the medial or basal portion.
